Monitoring the acylating reaction and extraction process of cefradine by HPLC

To analyze the concentration of 7-ADCA in acylating reaction mixture and residual cefradine in organic phase of extraction by HPLC to determine the reaction end piont. Chromatography conditions: column: Shim-pack CLC-ODS (5μm,150mm×6.0mm); mobile phase: methanol-phosphate buffer (pH7.0)-water (25∶5∶70); flow rate: acylating reaction 0.5ml/min (0~5min), 2.0ml/min (5~15min), extraction 1.5ml/min; detection wavelength: acylating reaction 263nm, extraction 254nm. The HPLC analytical procedures are sensitive, rapid and have greater specificity than most chemical or biological assays.
