Cellular Toxic Effect of Low Dose Cisplatin Combined with Interstitial Injection of ~(32)P Glass Microspheres on Mouse Solid Tumor S180

To investigate the cellular toxic effect of a combination of low dose cisplatin and 32P glass microsphere (32P-GMS) on mouse solid tumor S180. Twenty-eight mouse with solid tumor S180 were randomly divided into four groups (control group, cisplatin group, 32P-GMS group, combination of cisplatin and 32P-GMS group), and 7 for each group. After 2 weeks of treatment the weight of tumor was measured separately. The death rate of tumor cells and the inhibition rate of tumor were calculated respectively. The ultrastructural changes of specimens were observed by transmission electron microscopy. The inhibition rates in the three treatment groups were 47%, 40%, and 76%, respectively. There were significant differences in the inhibition rates between combination of cisplatin and 32P-GMS group and other groups (P﹤0.01). The death rate of tumor cells was 75% in the combination therapy group, which was significantly higher than those in other groups by observing under the electronic microscope. Most of tumor cells were disappeared which displayed a large area of necrosis structures. [Conclusion] The combination using of low dose cisplatin interstitial injection of 32P-GMS is able to enhance the cellular toxic effect on mouse solid tumor S180 comparing to single using of cisplatin or 32P-GMS .
