IN VITRO PURGING OF LEUKEMIC CELLS BY VERAPAMIL COMBINED WITH ADRIAMYCIN, VINCRISTINE AND ETOPOSIDE

Using semi solid culture technique of clonal formation in vitro, we observed the influence of verapamil (VPL) upon ex vivo purging activity of adriamycin(ADM), vincristine(VCR) or etoposide(VP 16). The results showed that VPL significantly enhanced the killing activity of ADM, VCR or VP 16 on leukemia progenitor (L CFU) cells, but did not add their toxicity to GM CFU. When purged by combination of 0.92 μmol/L ADM, 0 0250μmol/L VCR and 1.70μmol/L VP 16 or VPL combined with them, the survival rate of L CFU was 11.61%, 3.52%, respectively, with the survival rate of GM CFU was 35 81%, 26 05%, respectively, suggesting that VPL selectively enhanced ex vivo purging activity of combination of the above chemotherapeutic agents on leukemic cells, but did not add their toxicity to GM CFU.
