Screening and identification of the antagonistic organisms against Colletotrichum coccodes on potato

In this study,the biocontrol strain of Colletotrichum coccodes on potato was screened by confrontation culture method.The results showed that the strain ZHA10 had good inhibition against C.coccodes,and the inhibition rate reached 76.43%.In order to understand the bacteriostatic mechanism of the endophytic strain ZHA10 to C.coccodes and its taxonomic status,a series of physiological and biochemical experiments were conducted. The results showed that the antibacterial activity was mainly due to hyphal deformation and cracking.ZHA10 was short rod-shaped,gram-positive,and(0.5-0.75)μm×(1.5-3.1)μm in size,with endospores growing from the center to the tip.Through16S rDNA and gyrBgene sequence analysis,ZHA10 was identified as Bacillus subtilis. Besides,ZHA10 could effectively inhibit the growth of Alternaria solani,Fusarium oxysporum,Fusarium solani, Bipolaris sorokiniana,Fusarium oxysporumand Stysanus stemonitis,which showed that ZHA10,with broad spectrum and high antagonistic activity against pathogenic fungi,had good potential for development and application.
