Molecular detection and sequence analysis of Euphorbia leaf curl virus infecting Euphorbiain Shandong Province

In 2012,the diseased Euphorbia pulcherrima samples were collected from the Qingzhou flower market in Shandong.The samples was tested by PCR using degenerate primers(PA/PB)of geminivirus.The fragments of expected size(0.5 kb)were obtained and sequenced.The BLAST results showed that the causal agent of the disease was Euphorbia leaf curl virus(ELCV).The complete genome sequence of the isolate was then amplified. Analysis of similarity between the obtained complete genome sequence and those deposited in the GenBank database was performed.The results showed that this isolate(ELCV-Shandong,KC852148)shared a similarity of 99% with ELCV-G35(AJ558121)reported in Guangxi Province,and these two isolates were clustered into a small branch in the phylogenetic tree,and all Euphorbia leaf curl virus isolates reported in China were in the same branch.This is the first report that geminivirus infected Euphorbia pulcherrimain northern China.
