Zearalenone RIA was established and used to detect zearalenone in plants. Zeazalenone wa first converted into zearalenone-6'-carboxymethyloxime, and then identified by TLC, NRM and MS/MS, and conjugated with bovine serum albumin (BSA). The molecular ratio of zearalenone to BSA was about 23:1 as determined by UV scanning analysis. After five months of immunization of rabbit, the antibody titers reached 1:4×10~4. The cross reaction of this antiserum was very low (less than 0.05%) for phytohormones, but much higher for analogues of zearalenone, such as zearalanol. The Ab avidity wa 4.25×10~10 L/mol, and the detection limit was 3.5 pg. CV% of RIA in each sample batch and between sample batches was 6.1% and 8.6% respectively. Zearalenone in different plant samples was assayed using this method and the results indicated that zearalenone, as an endogenous active substance, occurred widely in higher plants.
