A Study on Genotoxicity of Four Kinds of Herbal Hair Dyes

MENG Zhen,CAI De-lei,SONG Song,et al.The Center for Disease Control and Prevention of Zhejiang Province,Hangzhou,Zhejiang,310051,China
Objective To understand the potential genotoxicity of four kinds of herbal hair dyes. Methods Ames test, chromosome aberration test in CHL cells and HGPRT locus forward mutation test in V79 cells were employed to investigate the effect of the four herbal hair dyes on gene mutation and chromosome aberration in both prokaryotic and eukaryotic cells in Vitro. Results An abnormal growth of reverse mutation colonies of TA97,TA98 and TA100 induced by sample 1 and 2 herbal hair dyes was observed, which were at the highest dose of 5000 μg/dish under S9mix activation system or without it.The same phenomenon was not occurred under the stimulation of sample 3 and 4 herbal hair dyes. There was no significant difference of chromosome aberration rate between the exposed and control cell(P 0.05),as well as the mutation frequencies at HGPRT locus.Negative data was also obtained in mammalian cells. Conclusion Sample 3 and 4 herbal hair dyes have no genotoxicity. It is difficult to draw a conclusions that sample 1 and 2 have no potential hazard, although the negative results were shown in assays based on mammalian system.
