Objective:to evaluate the value of lung resistance protein(LRP) and B lymphoma/leukemia 2(bcl 2) as predictors of resistance to chemotherapy in osteosarcomas.Methods:expression of LRP and bcl 2 was immunohistochemically studied in paraffin sections from 16 normal bone tissues and 42 specimens of osteosarcoma from patients who had not received any chemotherapy before surgery.Results:There was no detectable LRP or bcl 2 expression in 16 normal bone tissues.The positive expression rates of LRP and bcl 2 were 42 86%(18/42) and 52 38%(22/42) respectively and showed no significant correlation( P 0 05).Expression of LRP and bcl 2 had no relation with age,sex,histologic type and clinical stage( P 0 05).Evaluation for 27 osteosarcoma patients showed that difference in the response rates between the two groups was significant ( P 0 05).The predictive value of positive LRP stain for drug resistance was 85 7%(6/7),and that of negative stain was 55 0%(11/20).The response rate to chemotherapy of the bcl 2 positive group [13 3%(2/15)] was significantly lower than that of the LRP negative group[83 3%(10/12), P 0 01].The predictive value of positive bcl 2 stain for drug resistance was 86 7%(13/15) and that of negative stain was 55 0%.Conclusion:Detecting expression of LRP and bcl 2 may play an important role in judging drug resistence of human osteosarcoma.
