Discussion about the reaction condition in RAPD

This paper studied the influencing conditions of RAPD, including template extracting, template concentration, concentration of Mg 2+ and dNTPs, samples choosing and sample size. It concluded that high quality DNA template is vital in the reactions and its optimal dosage is 25～100ng; To obtain the optimal concentration of Mg 2+ and dNTP. The conditions of each primer and template should be optimized respectively. The result of amplification from population sample is better than from individuals and the numbers of samples do not affect the result obviously.
