Experimental study of the effect of compound shenqitang on nude mice with liver metastases

Objective:To establish a colonic adenocarcinoma liver metastases model for experimental study of the effect of compound shenqitang.Methods:Liver metastases were established by intrasplenic injection of colonic adenocarcinoma cells(Ls174t) with 1×10 6 cells in 0.1ml solution in 24 BALB/c nude mice which were divided into two groups randomly.Granular forage with compound shenqitang was fed in experimental group and granular forage without compound shenqitang was fed in control group daily. After three weeks every mouse was weighted and killed,blood of the mice should be collected .The weight,size and the number of the liver metastatic lesions were studied,every carcinoma node should be weighted,specimens were fixed by formalin and embedded by paraffin ,all slices with a thickness of 4μm were studied histologically.nm23H 1-NDPK expression in 24 nude mice with liver metastases from colon carcinoma was examined immunohistochemical (sp) by using antinm23.Using nested reverse transcript polymerase chain reaction(RT-PCR) technique CK20 mRNA in blood of 24 nude mice with liver metastasis from colon carcinoma was detected.Results:All mice inoculated with Ls174t cell lines developed liver metastases,other organic metastases weren't observed.The pathological results showed that tumour cells of liver metastases accorded with poorly differentiated human colonic adenocarcinoma . In experimental group the growth weight(4.12±0.53g) from experimental opening to ending was significantly higher than that(3.21±0.59g) in control group(P0.05). In experimental group the number(2.42±0.99n) and the weight(63.67±21.29mg) of liver metastases were significantly lower than those(4.17±1.99n,94.32±37.86mg) in control group(P0.05). In experimental group nm23H 1-NDPK positive expression rate (66.7%) significantly higher than that(16.7%) in control group(P0.05). Using semiquantitative examination, In experimental group the relative value of CK20 mRNA expression in blood (0.398±0.143) was significantly lower than that(0.518±0.090) in control group(P0.05).Conclusion:Compound shenqitang could effectively inhibit the development of liver metastasis carcinoma.
