Genetic Diversity Analysis of Lilium Based on psbA-trnH Gene Sequence Variation

Objective:Used chloroplast gene psbA-trnH sequence changes to study Lilium genetic diversity.Method:Lilium chloroplast psbA-trnH gene fragment was amplified by PCR and sequencing,and used biological software Clustalx2,Mega5.0 DnaSP to analysis sequencing results.Result:The amplified fragment length of PsbA-trnH gene was about 469 ~ 583bp,GC content of 34.47 to 35.20%.27 nucleotide variable sites,12 information sites.Variable sites were mainly concentrated in the 88-107bp region,Tested materials could be divided into three categories according to these variable sites,Nucleotide diversity was 0.01150.Conclusion:Chloroplast genes trnH-psbA sequences vriation could distinguish the difference between the Lilium species,it could be used as a supplement study genetic diversity method.
