Study on the Chemical Constituents and Antibacterial Activity of Radix Sophora Flavescens

Objective :To study the chemical composition and antimicrobial activity of Radix Sophorae flavescentis,and to use it for the add-on development. Methods :The root of Radix Sophorae flavescentis were isolated and purified from by macroporous resin column chromatography,silica gel column chromatography,Sephadex LH-20 column chromatography,reverse phase C_(18) open column chromatography and preparation of HPLC and other technical means. The drug sensitivity of the total alkaloids and their main alkaloids were investigated by the method of two times dilution and colony counting. Results :Sixteen compounds were isolated from Sophora flavescens,including 9 alkaloids and 7 flavonoids. The result shows that the total alkaloids of Sophora flavescens and its main alkaloid has certain antibacterial activity against Staphylococcus aureus,Escherichia coli,Pseudomonasaeruginosa,Beta hemolytic streptococcus and Candida albicans. The inhibitory effect of total alkaloids of the Radix Sophora flavescens was stronger than the individual compounds,and the antibacterial effect of Staphylococcus aureus and Candida albicans was better. The inhibitory effect of the individual compounds to Staphylococcus aureus was stronger than the other four strains. Inhibitory effects of matrine and sophocarpine to Staphylococcus aureus was stronger than its oxide. Conclusion :The total alkaloids and their main alkaloids of Radix Sophorae flavescentis have obvious antimicrobial effect,and the antibacterial activity of each monomer was different,which could be used as the basis for the development of two times.
