Preparation and Characterization of GHK-Cu Liposome and its Effects on Hair Growth in Mouse Alopecia Model

Objective To prepare GHK-Cu liposome and analyze its effect on hair growth in mouse alopecia model.Methods Reverse evaporation method was used for GHK-Cu liposome preparation.The alopecia model was established by spreading 6% of Na2S solution on the back of KM mice.The mice were divided into three groups randomly and treated with 0.6 mol·L-1 of GHK-Cu,0.6 mol·L-1 of GHK-Cu liposome,or distilled water once daily,respectively.The new hair growth status was recorded every day.Paraffin sections were obtained from treated skin tissues post treatment,and the pathological and histological changes were evaluated.Western blotting was carried out to detect VEGF expression in the three groups.Results Both of GHK-Cu liposome and GHK-Cu could promote hair growth in the mouse alopecia model.The number of hair follicles in the two groups was significantly different from that of the control group.VEGF expression in GHK-Cu and GHK-Cu liposome groups was higher than that in the control group before the hair grew completely.Conclusion Liposome can effectively encapsulate GHK-Cu,and maitain the function of GHK-Cu in promoting hair grow.Both GHK-Cu liposome and GHK-Cu can increase VEGF expression in mouse skin at early stage of hair growth.
